Product Introduction
Bovine serum is the most used natural medium for cell culture. It is rich in nutrients necessary for cell growth. It is often used for in vitro culture of animal cells. It has extremely important functions: (1) Provides hormones for maintaining exponential cell growth. There are no or very few nutrients in the medium, as well as the main low-molecular nutrients. (2) Provide binding proteins, which can recognize vitamins, lipids, metals and other hormones, and can bind or modulate the vitality of the substances they bind. (3) In some cases, the binding protein can combine with toxic metals and pyrogens to detoxify. (4) It is a source of factors needed for cells to adhere to and spread on the plastic culture medium. (5) Act as a pH buffer. (6) Provide protease inhibitors to inactivate the remaining trypsin during cell passage and protect cells from harm.
According to the bovine birth time and the method of serum collection and separation, serum is generally divided into fetal bovine serum, newborn bovine serum and calf serum. Fetal bovine serum is made by puncturing the heart of eight-month-old fetal bovine blood, then heat inactivating, dialysis, and filtration sterilization. This product is a standard fetal bovine serum, which has been heat-inactivated and filtered to sterilize. It does not need to be heat-inactivated before use. It can be used after slow thawing at low temperature.
Storage and transportation
Transported on dry ice; stored at -20°C, valid for 5 years.
Instructions for use
1. Thawing: A gentle heating process can effectively reduce the generation of precipitation. It is not recommended to thaw serum directly at high temperature.
1) Move the frozen serum to a refrigerator at 2-8°C and thaw overnight;
2) Or move the serum to a 37°C water bath, shake and mix the dissolved substance at the bottom intermittently until all is dissolved.
2. The concentration of fetal bovine serum in the basic medium is generally 5-20%, and it is added in proportion to the cell types and experimental needs.
Precautions
1. Avoid repeated freezing and thawing. After the serum is thawed, it can be stored at 2-8°C for 6 weeks. If the storage time of the thawed serum needs to be longer than 6 weeks, it is recommended to divide it into a suitable volume and re-freeze it.
2. If the serum contains a large amount of precipitate, it is not recommended to filter the serum directly. If there is a need for filtration, the serum can be aliquoted into sterile centrifuge tubes, centrifuged slightly, and the supernatant is directly added to the culture medium and mixed before filtering.
3. If the serum is not mixed and placed in a water bath higher than 40°C, the protein at the bottom will be denatured to form a precipitate.
4. Pay attention to aseptic operation during use to prevent bacterial contamination.
5. Please wear lab coat and disposable gloves during operation.
